Isolasi Dan Uji Toksisitas Senyawa Metabolit Sekunder Fraksi Kloroform Ph 12 Dari Kulit Batang Pulai Basung Alstonia Spatulata
SUMMARY
Pulai basung (Alstonia spatulata Bl.) is one of medicinal plants from Alstonia genus and Apocynaceae family. The root, stem bark, leaves and fruits of this genus had been used in traditional medicine to alleviate some diseases. The goals of this study were to isolate and characterize secondary metabolites from chloroform fraction at pH 12 of stem bark pulai basung and analyze their toxicity by brine shrimp lethality test method. In this study the acid-base extraction method was used. The sample was extracted with maceration method using methanol solvent, then partitioned using chloroform. The fraction of chloroform at pH 12 was separated using Flash Chromatography Column obtained 13 fractions. The fractions were subjected to using TLC and obtained brown caramels with weight of 2,4 mg. Purity of the compound was analyzed using thin layer chromatography (TLC) and high performance liquid chromatography (HPLC). The compound was coded ASP-K12-FL-V32 and characterization using UV-Vis and FTIR spectrophotometer. The UV-Vis spectra showed a maximum absorption at wavelength of 243,5 nm, meanwhile FTIR spectra showed that the compound ASP-K12-FL-V32 possessed some bands at of 3573 cm-1 (NH), 3329 cm-1 (OH), 2926 cm-1, 2869 cm-1, and 2849 cm-1 (aliphatic C-H), 1733 cm-1 (C=O), 1564 cm-1 and 1456 cm-1 (aromatic C=C), and 1024 cm-1 (C-O). The results of the toxicity test using the brine shrimp lethality test (BSLT) method showed that the chloroform pH 12 fraction was toxic with LC50 value of 122,72 ppm. This concluded that ASP-K12-FL-V32 might be an alkaloid and it was considered to be moderate toxic.
