Isolasi Dan Uji Toksisitas Metabolit Sekunder Ekstrak N-Heksana Daun Benalu Pohon Karet (Helixanthera Sp)
Rubber tree mistletoe (Helixanthera sp) is a plant from the Loranthaceae family. This plant is still considered a parasite by local people and is not used as a raw material for medicines because local people do not know the properties of this mistletoe. Therefore, researchers conducted research on the Helixanthera sp species with the aim of isolating and characterizing secondary metabolite compounds from n-hexane extract of rubber tree mistletoe leaves (Helixanthera sp) and carrying out toxicity tests using the Brine Shrimp Lethality Test (BSLT) method. Isolation of secondary metabolite compounds in rubber tree mistletoe plants (Helixanthera sp) using gravity column chromatography to separate the compounds and obtained 4 fractions. Of all the fractions, the fraction that was further isolated was fraction 2. Fraction 2 was recrystallized and a white solid of 0,0773 g was obtained which was coded HS-H-F2. The HS-H-F2 compound was tested for purity using TLC with three different eluents and a melting point test using a Fisher Johns melting point determination tool. The HS-H-F2 compound was declared pure because there was one spot on the TLC plate for each eluent and the melting point test results carried out on the HS-H-F2 compound had a melting point of 168oC-170oC. Characterization of the HS-H-F2 compound was carried out using a UV-Vis and FTIR spectrophotometer. Characterization of the compound using a UV-Vis spectrophotometer showed the absence of conjugated phi (π) bonds at a wavelength of 200-400 nm, while the IR spectrum results showed the presence of wave numbers at absorption of 3199 cm-1 (O-H), 2944-2853 cm-1 (C-H aliphatic) and 1034 cm-1 (C-O). Based on the characterization results, the HS-H-F2 compound is a secondary metabolite compound in the terpenoid group. The results of the toxicity test carried out using the BSLT method showed that the HS-H-F2 compound was non-toxic with an LC50 value > 1000 ppm.
