ISOLASI DAN UJI ANTIOKSIDAN METABOLIT SEKUNDER DARI EKSTRAK DIKLOROMETANA TUMBUHAN KELAKAI (Stenochlaena palustris)
Stenochlaena palustris or often called kelakai is a fern that found in the tropical and subtropical forest and it thrives on peat soils also found to grow well in sandy soils. Kelakai are widely use as vegetable and traditional medicine and are also known as natural antioxidant sources. The aim of this research was to isolate the secondary metabolics of S. palustris and to determine the antioxidant activity of its extracts. Isolation process starts with extraction used maceration method and the extract from maceration was partitioned by solvents of n-hexane, dichloromethane and ethyl acetate. Antioxidant activity was tested using the DPPH method and ascorbic acid used as a positive control. The dichloromethane extract showed strong activity with values of IC50 54,91 µg/mL (AAI =1,45) and separated using Vacuum Liquid Chromatography (VLC) and the result of VLC was 13 fractions. The fraction F9 showed moderate activity with values of IC50 161,55 µg/mL (AAI= 0,49) and separated using Flash Chromatography and the result was 171 vials. Vials of 70-79 showed the formed crystal was tested by TLC and obtained the yellow crystal with the melting point of 166-168ºC, crystal was labelled as 19-04-B-FIX-SF71-79/171. Analyze of FTIR shown the absorption in wavenumber 3329,28 cm-1 (O-H), 2932,89 cm-1 (C-H aliphatic), 1733,12 cm-1 (C=O), 1648,24 cm-1 (C=N), and 1249,93 cm-1 (C-O). The antioxidant test result of 19-04-B-FIX-SF71-79/171 was values of IC50 >1000 µg/mL (AAI= 1,09 x 10-3) and it indicates the antioxidant activity of the crystal was very weak.
