Live microorganisms that have potential as probiotics need to be evaluated for their safety and antioxidant activity to qualify as probiotics. The purpose of this studywas to evaluatethe safety and antioxidant activity of Lactobacillus fermentum InaCC B1295 encapsulated in cellulose microfiber (CMF) hydrogel from oil palm trunk during storage. This study was conducted experimentally, consisting of safety evaluation analyzed descriptively and antioxidant activity analyzed statistically in a factorial complete randomized design. The experimental unit consisted of two factors, namely storage temperature (27‒30C and 4C) and storage duration (0, 14 and 28 days) with three replications each, resulting in 18 experimental units. Parameters observed in this study were safety evaluation (hemolytic activity, cytolysin and gelatinase production), antioxidant activity consisting of DPPH radical capture ability and hydroxyl radical scavenging (HRS). The observed data of antioxidant activity were analyzed for variance and continued with the duncan multiple range test at 5% level. The results of safety evaluation observations show that Lactobacillus fermentum InaCC B1295 encapsulated with CMF is classified as safe, which is characterized by the absence of hemolytic activity and the production of cytolysin and gelatinase by this. The storage temperature, storage time, and their interactions significantly affected antioxidant activity of DPPH radicals ranging from 87.30 to 189.35 ppm. Additionally, the results also revealed that storage time, but not storage temperature and interaction significantly (P0.05) influenced HRS value. The CMF encapsulated LAB from OPEFB was shown to have good antioxidant activity, be safe to eat, and have potential probiotic uses based on these studies.
