Skrining Kemampuan Jamur Amanita Sp. Dan Lactarius Sp. Isolat Taman Nasional Sebangau, Kalimantan Tengah Sebagai Penghasil Lakase
Laccase is an enzyme belonging to the multicopper oxidase group, capable of catalyzing various substrates, including phenolic and non-phenolic compounds. Laccase is considered as an ideal "green catalyst" due to its catalytic capabilities, making it widely used in industrial processes. The most commonly applied laccases in biotechnology are mostly derived from fungi. White-rot fungi from the Basidiomycota are known to be the highest producers of laccase. This study aims to qualitatively determine the potential of Amanita sp. (sect. Vaginatae) M15 and Lactarius sp. M35 as laccase-producing fungi using ABTS and guaiacol substrates. The method used for screening laccase-producing fungi was by growing fungal mycelial plugs on PDA medium at pH 4.5 and 5.5, which had been supplemented with laccase indicator substrates (guaiacol or 2,2'-azino-di-3-ethylbenzothiazoline-6-sulfonate, ABTS). The sterilized agar media were supplemented with guaiacol or ABTS solution, which was filter-sterilized using a 0.45 µm syringe filter. Mycelial plugs (±1 cm), previously grown, were implanted in the center of the screening media using a cork borer. Petri dishes were incubated at 30°C and observed for up to 12 days. Laccase activity was indicated by the formation of reddish-brown halo for the guaiacol substrate and dark blue-green (teal) halo for the ABTS substrate. The addition of CuSO4 was intended to enhance laccase activity, as copper ions are part of the active site of laccase involved in electron transfer. The research results show that Amanita sp. (sect. Vaginatae) M15 and Lactarius sp. M35 exhibit a color change to brownish-red with both indicators (guaiacol and ABTS) at different pH levels, with and without the addition of CuSO4. The reddish-brown color observed in both fungi is most likely a pigment secreted into the medium by the fungi when xenobiotic compounds such as guaiacol or ABTS are added. Both fungi produced a darker brown pigment at pH 4.5 compared to pH 5.5.
