Karakterisasi Parsial Dan Aktivitas Enzim Hidrolitik Bakteri Endofit Paku Lau (Acrostichum Aureum L.) Dari Pulau Bengkalis
Endophytic bacteria are bacteria that can be found in plant tissue and have mutualistic symbiosis without causing disease symptoms or damaging plant tissue. Endophytic bacteria are known to produce enzymes, antibiotics and others. This study aims to determine the characteristics of 25 isolates of endophytic bacteria from sea fern (Acrostichum aureum L.) macroscopically, physiologically and biochemically and to test the potential of bacteria as producers of hydrolytic enzymes, namely amylase, cellulase and lipase. Media to see the activity of the amylase enzyme with 2% starch content, cellulase enzyme with 1% CMC and lipase enzyme with 2% olive oil substrate with phenol red indicator. The results of the study obtained morphology as much as 48% have milky white colonies, 36% yellowish white, 8% red and 4% yellowish. 96% have a circular shape and 4% irregular. 96% have entire edges and 4% undulate. 56% have raised elevation, 32% convex and 12% flat. In the physiological observation of the environment (temperature 4˚C, 28˚C, 45˚C, salt concentration 1%, 1.5%, 2% and pH 5, 7, 9) of 25 endophytic bacterial isolates at a temperature of 4˚C all bacteria did not grow. At a temperature of 28˚C, all endophytic bacterial isolates were obtained to grow well. At a temperature of 45˚C, 7 isolates were obtained to grow well. Then for the test of salt concentrations of 1%, 1.5% and 2%, all isolates were obtained to grow well because they were still within the range that supports bacterial growth. Furthermore, in the pH test, at pH 5, 20 isolates were obtained to grow well. At pH 7 and pH 9, all isolates were obtained to grow well. Furthermore, in the biochemical test (methyl red and catalase), 23 isolates were obtained to show positive results for endophytic bacteria indicating that the bacteria were able to ferment glucose through a mixed fermentation pathway. Then, the 3% catalase test showed positive results for all endophytic bacteria. In the amylase activity test, there were three isolates with amylase activity, seven isolates with cellulase activity and six isolates with lipase activity. Two isolates were obtained that were able to produce two enzymes, namely isolate B.S.B 2.4 in the amylase and lipase enzymes and B.S.B 2.7 in the amylase and cellulase enzyme tests.
