Induksi Tunas Pisang Kepok (Musa Paradisiaca L.) Menggunakan Bap Pada Media Ms Cair Secara In Vitro
The propagation of pisang kepok using seedlings derived from its parents is an obstacle for farmers to produce large scale and quality seedlings. In vitro culture is a solution for banana seedling propagation. The growth rate of plants can be supported by the provision of cytokinin type growth regulators (ZPT) such as 6 Benzyl Amino Purin (BAP). This study aims to determine the effect of BAP on shoot propagation from pisang kepok (Musa paradisiaca L.) explants in vitro and determine the effective concentration of BAP for shoot propagation. This study used a completely randomized design (CRD) with 7 (seven) treatments levels repeated 5 (five) times resulting in 35 experimental units. The treatments consisted of control (0 mg/L BAP) , 1 mg/L BAP, 2 mg/L BAP, 3 mg/L BAP, 4 mg/L BAP, 5 mg/L BAP, and 6 mg/L BAP. The parameters observed were the percentage of living explants, percentage of nodules formed, and time appeared nodules .Based on the results of this study, percentage of living explants in banana stem explants given concentrations of 0,1, 2, 3, 4, 5 and 6 mg/L BAP ranged from 80% to 100%. The percentage of nodule formation, nodule emergence time and number of nodules from kepok banana stem explants were found in the 3 mg/L BAP treatment, respectively, namely 20%, 65th day after planting and one nodule.
