Ekstraksi Dan Karakterisasi Ekstrak Kasar Enzim Pepsin Dari Lambung Ikan Lele (Clarias sp.)
Fish stomach is a by-product of the fishing industry that has the potential to be developed as a source of pepsin enzyme, for example from catfish stomach. The objectiveof thisstudy wasto determinethecharacterization of crudepepsin enzyme from catfish stomach. The pepsin extraction process began with separating the stomach wall fluid from the fish by adding Tris-HCl buffer at pH 7.5, followed by activating pepsinogen using HCl at pH 2. The results of the study indicated that pepsin enzyme can be extracted from catfish stomach and had an enzyme activity of 33.500±0.866 U/mL, a protein concentration of 0.358±0.005 mg/mL, total enzyme activity of 1608±41.569 U, total protein of 17.192±0.220 mg, and specific activity of 93.523 U/mg. The characteristics of the crude pepsin enzyme extract from catfish stomach showed optimal activity at 50°C and pH 4, while the relative activity of pepsin enzyme toward metal ions NaCl, ZnCl₂, and FeCl₃ was 81.081±1.560%, 49.099±1.440%, and 128.153±1.460%, respectively.
