Isolasi Dan Identifikasi Bakteri Asam Laktat Dari Dadih Asal Kabupaten Kampar Sebagai Pangan Fermentasi Tradisional
ABSTRACT
Lactic acid bacteria (LAB) have become the object of extensive research,
both nationally and internationally, because this bacterium has a critical role in
most fermented foods. This research aims to isolate and identify the presence of
LAB in dadih from Limau Manis Village. The research was conducted by
isolating LAB from two tubes of dadih using the serial dilution method. Each
dilution result was inoculated four times on a petri dish using the spread plate
method. The colonies that grew from the spread plate results were observed for
their morphology and recorded. The next stage was purification by streak plate
method to obtain single colony and inoculation of the isolated LAB into the broth
media for further testing. The data obtained were tabulated and analyzed
descriptively. This research resulted in 15 isolates finding. Gram staining shows
14 blue isolates and one rod shaped isolate with red colour. Catalase test shows
negative result for 14 isolates except LM 2.1 isolate. About 11 isolates produce
gas and 4 isolates do not. Based on all test results, 14 isolates are expected to be
LAB with the genus Lactobacillus or Bifidobacterium.
