Education plays a crucial role in advancing a nation to be able to adapt to the demands of 21st-century education, which requires students to master science process skills. Learning biology involves direct observation of the objects under study through laboratory or field practical activities. Biology practicum requires a guide that is designed in alignment with the curriculum and the demands of the 21st century to help develop students’ science process skills. In the learning process, there are various obstacles faced by both teachers and students. These obstacles are among the factors that contribute to suboptimal learning outcomes. One of the main issues is the lack of students’ science process skills, which leads to less meaningful learning experiences.
As a solution, the researcher proposed an innovation by developing a learning material in the form of a practicum guide based on guided inquiry to enhance students’ science process skills. The main topic developed in the practicum guide is the human circulatory system for Grade XI Senior High School Biology. This study consisted of two stages: the first stage was the development of the guided inquiry-based practicum guide using Research & Development (R&D) through the ADDIE approach (Analysis, Design, Development, Implementation, and Evaluation) conducted in the Master’s Program of FKIP UNRI; the second stage was an experimental implementation at MAN 1 Indragiri Hulu.
The development phase included a validity test involving three experts (a subject matter expert, an education expert, and a media expert) who assessed the validity using validation sheets. The practicality test involved two teachers from MAN 1 Indragiri Hulu. The practicality of the guided inquiry-based practicum guide was evaluated through response questionnaires, teacher practicality tests, one-on-one trials, and small-scale trials. In the experimental phase, the impact of the guided inquiry-based practicum guide on students’ science process skills in the human circulatory system topic was examined. The assessment of students’ science process skills was carried out using multiple-choice pretest-posttest questions, observation sheets, and worksheets embedded in the practicum guide.
The validity test results from the material, education, and media experts indicated that the guided inquiry-based practicum guide received a score of 3.78, categorized as very valid. The practicality test results showed an average score of 3.95, categorized as very practical. The experimental results demonstrated a significant influence of the guided inquiry-based practicum guide on students’ science process skills. In the control class, the average score was 73.3 (good category) with an N-gain of 0.5 (moderate category), while in the experimental class, the average score was 81.48 (very good category) with an N-gain of 0.7 (high category).

Based on the results of the development and implementation tests, it can be concluded that the guided inquiry-based practicum guide on the human circulatory system topic is significantly more effective in improving students’ science process skills in the experimental class compared to the control class.
