Identifikasi Senyawa Bioaktif Dan Uji Aktivitas Antikolesterol Dari Fraksi Teripang Keling (Holothuria Atra) Secara In-Vitro
Holothuria atra is a type of sea cucumber that has high ecological and economic value. Holothuria atra is known to contain various secondary metabolites such as alkaloids, flavonoids, steroids/terpenoids, saponins, and phenolics. These compounds have great potential in the food and pharmaceutical industries because they play arolein improving human health. This studyaims to identify the bioactive compounds in H. atra fractions that have potential as anticholesterol agents. The study uses experimental methods with direct analysis in the laboratory. The parameters observed included flour and extract yields, moisture and fat content tests, qualitative and quantitative secondary metabolite tests, antioxidant activity, and anticholesterol activity tests of the sea cucumber fraction. The results showed that the flour yield was 3.27%, and the crude methanol extract was 20.00%. The moisture and fat contents were 12.46% and 2.39%, respectively. The results of secondary metabolite identification showed the presence of bioactive compounds in the form of saponins and triterpenoids, which were dominant in the n-hexane and ethanol fractions, while the ethyl acetate fraction was only detected positive for triterpenoids. No alkaloids, flavonoids, or phenoliccompounds were detected in the three fractions. Quantitative secondary metabolite testing of the n-hexane fraction of H. atra produced a saponin content of 0.029%, while the ethanol fraction produced saponin of 0.173%. Antioxidant activity based on IC₅₀ values was 359.66 ppm for n-hexane, 555.02 ppm for ethyl acetate, and 292.57 ppm for ethanol. The anticholesterol activity of each fraction was n-hexane 41.60%, ethyl acetate 36.51%, and ethanol 47.71%. Based on these results, the ethanol fraction showed the strongest antioxidant activity and had the highest anticholesterol potential compared to the other fractions.
