Perbandingan Ekspresi Gen Interleukin-8 Dan Transforming Growth Factor-Β Pada Sel Punca Mesenkimal Dengan Suplementasi Human Platelet Lysate Dan Fetal Bovine Serum
Mesenchymal stem cells (MSCs) have great potential in regenerative therapy, but conventional culture using fetal bovine serum (FBS) poses biological and ethical risks, making human platelet lysate (HPL) a safer alternative. This study aims to compare the expression of interleukin-8 (IL-8) and transforming growth factor-β (TGF-β) genes in MSCs cultured using HPL and FBS supplementation. This experimental study uses a post-test only control group design. Mesenchymal stem cells were isolated from Wharton's Jelly of the umbilical cord and cultured to the third passage before being divided into two groups: 5% HPL culture and 10% FBS culture (n=6 each). IL-8 and TGF-β mRNA expression was measured using RT- qPCR and analyzed using the ΔΔCt method, followed by statistical analysis using the Shapiro–Wilk test and independent t-test. The study results showed that IL-8 and TGF-β expression in the FBS group was slightly higher than in the HPL group (IL- 8: 1.024 vs. 0.920; TGF-β: 1.106 vs. 0.946), but there was no significant difference (p>0.05).
