Pengaruh Tingkat Kematangan Embrio Dan Kombinasi Ga3 Dengan Naa Pada Media Ms Terhadap Kultur In Vitro Embrio Kelapa Sawit (Elaeis Guineensis Jacq.)
ABSTRACT
The propagation of oil palm (Elaeis guineensis Jacq.) for the provision of
superior seedlings faces challenges that can be addressed through tissue culture.
The success of this technique is highly dependent on factors such as the explant
type and the use of plant growth regulators (PGRs). This study aimed to investigate
the interaction and main effects, as well as to determine the optimal combination of
embryo maturity (immature and mature) and the composition of PGRs Gibberellin
(GA3) and naphthaleneacetic acid (NAA) in MS media on the in-vitro growth of oil
palm embryo explants. The research was conducted from February to May 2025,
the study used a factorial Completely Randomized Design (CRD) with two factors.
The first factor was embryo maturity (immature and mature embryos). The second
factor was the PGR composition with five levels (including a control without PGRs
and various combinations of GA3 and NAA). Data were analyzed using Analysis of
Variance (ANOVA) and followed by Duncan's New Multiple Range Test
(DNMRT) at the 5 % significance level. The results indicated that the interaction
between embryo maturity and PGR composition had a significant effect on all
observed parameters. The interaction of immature embryos with 1 mg/l GA3 was
the best treatment for plumule emergence time (11.35 DAP), radicle emergence
time (55.56 DAP), and explant survival rate (93.3 %). Meanwhile, the best
treatment for plumule length (4.46 cm), radicle length (1.53 cm), number of leaves
(1.47), and number of roots (1.87) was obtained from the interaction of immature
embryos with 1 mg/l GA3 + 5 mg/l NAA.
