Amplifikasi Gen Housekeeping Beta-Tubulin Pada Tumbuhan Tuntun Angin (Elaeocarpus Floribundus) Dari Danau Kajuik Kabupaten Pelalawan Provinsi Riau
Tuntun Angin plants are plants that grow around the river exposure to floods in Lake Kajuik, Pelalawan Regency, Riau Province. Tuntun Angin plants can be used as material to study the tolerance mechanism of flooding stress. Analysis of gene expression requires internal controls. One example of internal controls is Beta-tubulin that is a member of housekeeping genes. This study aims to amplificate the beta-tubulin gene on Tuntun Angin from Kajuik Lake, Pelalawan Regency, Riau Province. The research methods were leaves collection, DNA isolation, polymerase chain reaction (PCR), electrophoresis, sequencing, data analysis using BLASTn, MEGA and Primer3 programs. The results showed that total DNA of Elaeocarpus floribundus has been obtained and proved by amplification of matK gene. Tuntun Angin of beta-tubulin gene has also been obtained with the size of 810 bp. Tuntun Angin beta-tubulin DNA sequence had similarity of around 74-78% to some plant species. The isolated beta-tubulin gene was the first reported from the Elaeocarpus floribundus species. The beta-tubulin primers spesifically for Tuntun Angin (Elaeocarpus floribundus BI) have been designed such as Forward 5'-AGATCACCCACTCTTGG-3' and Reverse 5'-AAGGTGGATGACATCTTGAG-3'
