Aktivitas Kandidiasis Dari Metabolit Sekunder Penicillium Sp. Lbkurcc34
Penicillium sp. LBKURCC34 is a fungi isolated from Primary Forest Peat Soil Biosphere Reserve in Giam Siak Kecil Bukit Batu (GSKBB) Province of Riau. This fungus has the ability to produce chitinase and antimicrobial activity. Candidiasis is an infectious disease caused by Candida albicans. The purpose of this study to separate the crude extract of secondary metabolites from the growth media of Penicillium sp. LBKURCC34 which has antifungi activity of Candida albicans. Production of secondary metabolites was carried out by fermentation in liquid media which was inoculated with Penicillium sp. spores LBKURCC34 (7 x 1012 spores for 50 mL media). Fermentation was carried out for 14 days and extracted with distilled ethyl acetate solvent. Ethyl acetate extract was evaporated, then separated by flash column chromatography. Antifungal tests were carried out by disc diffusion method on the fungi Candida albicans with a concentration of 5.7 mg/mL. Amoxicillin and ketoconazole were used as positive controls and methanol as solvent controls. Further identification of crude extract was tested by TLC by comparing the eluent ethyl acetate: n-hexane (1:1; 3:2; and 7:3) below UV 254 nm and 366 nm. HPLC analysis crude extract produced 7 peaks in a chromatogram with a wavelength of 210 nm and 5 peaks in a 254 nm wavelength chromatogram. The fraction of column chromatography produced 13 fractions. In the antifungal test, from the fractions of 1-13 which showed the activity of fraction 2 (F2), fraction 3 (F3) and fraction 4 (F4) which was characterized by the presence of a clear zone.Then the best antifungal activity is found in fraction 3.
