Isolasi Gen Housekeeping Gapdh (Gliseraldehid-3-Fosfat Dehidrogenase) Pada Tumbuhan Sirsak (Annona Muricata L.)
Soursop plant (Annona muricata L.) is one of the plants in Indonesia that has many benefits. One of them as a medicinal plant. This ability is related to certain genes that encode certain proteins. Initial studies were conducted to isolate the housekeeping gene, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), which can act as an internal control. The research procedure begins with the provision of samples, total DNA isolation, electrophoresis, PCR, sequencing and data analysis. Data analysis used the BioEdit version 7.0, BLASTn, MEGA version 7.0 and Primary3 programs. The soursop sample used was correct from the soursop plant (Annona muricata L.) based on matK gene analysis which has a size of 626 bp. The GAPDH gene has also been isolated with a size of 961 bp with 5 exons and 5 introns. A specific GAPDH primer pair for soursop plants have been designed (Annona muricata L.) namely forward primer with sequence of 5'-GGT TTG GCA TTG TTG AGG GT-3’ and reverse primer with sequence of 5'-TCA ACG GTA GGA ACA CGG AA-3’ with PCR product measuring 465 pb.
