Kultivasi Mikroalga Menggunakan Media Air Gambut Berdasarkan Perbedaan Fotoperiode Dan Penambahan Jumlah Kno3
Microalgae cultivation is influenced by several factors, including nutrition on the media, light, and CO2. In this research, peat water is used as a cultivation medium, the presence of nutrients contained in peat water has the potential to be used as a microalgae cultivation medium. Therefore, this research has the aim of determining the effect of photoperiods and adding different amounts of KNO3 to optical density, growth rates and lipid content of microalgae cultivated on peat water media. The stages carried out in this study were sterilization of tools with an autoclave of 121˚C for 15 minutes at a pressure of 2 atm to avoid contamination of other organisms, the preparation of peat water, microalgae precultures with a ratio of 95 ml of peat water plus 5 ml of Chlorella sp. microalgae is carried out for 10 days, and the cultivation process of microalgae is carried out by inserting 10 ml of algal samples from the pre-culture stage into the pre-culture stage. 90 ml of peat water. Fixed variables used were aeration rate of 2.3 L / min, light intensity 1500 lux, free variables of photoperiod 12 T: 12 G, 16 T: 8 G and 24 T : 0 G and the addition of KNO3 nutrients by 0.25 g / L, 0.5 g / L, 0.75 g / L and, 1 g / L. The results showed that the highest optical density value is 1.0718 was obtained at 16 T: 8 G photoperiod with the addition of 0.75 g / L KNO3. The value of the growth rate of 0.2473 / day at 24 T: 0 G photoperiod with the addition of 1 g / L KNO3. The highest lipid content was obtained at 24 T: 0 G photoperiods at 37.39%, with the addition of 0.25 g / L KNO3 nutrition.
