Seleksi Dan Analisis Keragaman Genetik Plasma Nutfah Kelapa Sawit (Elaeis Guineensis Jacq.) Di Pt Smart Tbk Berdasarkan Marka Snap Berbasis Gen Stearoyl Acid Desaturase
ABSTRACT
The research aims were to determine the SNAP (Single Nucleotide Amplified Polymorphism) marker based on the SAD (Stearoyl acid Desaturase) gene for oil palm genetic diversity analysis and obtain commercial oil palm candidates that have high oleic acid content. The SNAP primers were designed based on SNP position on SAD genes. All of 42 SNAP primers were subjected to optimum annealing temperature analysis using 3 oil palm DNA sampels which wre E. guineensis, E. oleifera and Eo x Eg hybrids The annealing temperature was ranged from 540C – 620C. Eight (8) primers were selected for amplification of 42 oil palm samples. The amplicon was scored based on the SNP alleles. SNP alleles were converted to numeric based on software requirement. Oil palm genetic diversity was analyzed using Darwin software, Polymorphic Information Content (PIC) was analyzed using Cervus software and SNAP primer characteristics were analyzed using Genetic Analysis in excel (GeneAlex) software. The results showed that there were variations in the population tested based on SNAP markers used The average PIC of the SNAP primer was 0.52. Genetic similarity analysis showed that oil palm samples were divided into two groups. The first group were consisted of 24 samples from Spear, Evo, Tan, Dura, Com, BTC, Gha and Eko population, whereas the second group consisted of 18 samples from Gha, Com, Nig, ET, spear, Dura, BTC, Eko and Evo population. Only ET and Nig samples were consisted of all their individual samples in second groups, and the other populations were separated into two groups. There is no individual E. guineensis samples was indicated has high oleic acid content.
