Optimalisasi Waktu, Kadar Air Dan Konsentrasi Cu2+ Fermentasi Padat Batang Padi Dan Serbuk Gergaji Produksi Lakase Trichoderma Asperellum Lbkurcc1
Laccases (benzenediol: oxygen oxireductase, EC 1.10.3.2) are
metaloenzymes that can catalyze the oxidation reaction of phenol compounds,
amina and non-phenolic compounds. Laccase can be produced by Trichoderma
asperellum LBKURCC1 through submerged fermentation with rice husk
substrate, but with low activity. Production of laccase can be increased by using
different fermentation methods, such as Solid State Fermentation (SSF), and by
optimising fermentation conditions. This study aims to optimize the SSF
production of laccase by T. asperellum LBKURCC1 through optimisation of three
parameters, namely fermentation time (X1), water content (X2) and concentration
of Cu2+ as CuSO4.7H2O (X3), using rice straw and Dipterocarpus sp. sawdust as
inducer. Enzyme extraction was carried out by adding 20 mL 0.05 M sodium
acetate buffer (pH 5.5) to 2 grams of substrate at the end of fermentation. Laccase
activity was measured using 2,2’-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)
(ABTS) as substrate. Optimization of laccase production was evaluated by the
Central Composite Design (CCD) and Response Surface Methodology (RSM)
using Minitab program version 17.1. ANOVA results indicate that the quadratic
model was the chosen model (p-value lack-of-fit> 0.05). The three variables
