Isolasi Dan Uji Penghambatan Α-Glukosidase Senyawa Metabolit Sekunder Dari Ekstrak Paku Kelakai (Stenochlaena Palustris)
Stenochlaena palustris is one of the ferns that are often found in peat, and it has
medicinal properties including fever, immunostimulant, diarrhea, and antiaging.
According to a published paper, water extract from this species posses
α-glucosidase inhibitor. However, the secondary metabolites underliying this
activity has not yet been identified. Thus the objectives of this study were to
determine the potential the activity α-glucosidase inhibitor from various extract
and to isolate their secondary metabolites from the species. Air-dried aerial parts
of the species were extracted by using maceration in metanol and followed by
liquid-liquid extraction against n-hexane, dichloromethane (DCM) and ethyl
acetate to obtain n-hexane, dichloromethane, ethyl acetate and water extracts. The
α-glucosidase inhibitor activity of these extract revelead that DCM extract possess
the activity with IC50 value 406.26 μg/mL compared to n-hexane extract, ethyl
acetate and water. This extract is then purified through the chromatography
technique Vacum Liquid Chromatography (VLC) and yielded thirteen fractions.
In fraction 8 found solids that have the potential to produce crystals so it is
necessary to recrystallize to obtain crystals. The crystals (19-04-B-F8) obtained
had a melting point of 172-173 OC and phytochemical analysis revealed that the
compounds obtained belonged to the flavonoid group. Fourier-tranform infrared
spectroscopy (FTIR) analysis depicted the presence of absorption band OH at
3226 cm-1, absorption band C-H alkene at 3026 cm-1, absorption band C-H alkane
at 2890 cm-1, absorption band OCH3 at 2814 cm-1, absorption band C=O at 1696
cm-1, absorption band C=C at 1603 cm-1, absorption band C-H alkane at 1362
cm-1, absorption band C-O at 1261 cm-1, 1205 cm-1, 1179 cm-1, 1171 cm-1, 1085
cm-1, dan 1063 cm-1 dan absorption band C-H aromatic at 827 cm-1. The fraction
and the compound exhibited a range of inhibitory activities against α-glucosidase
with the compound (19-04-B-F8) exhibiting an IC50 value of 301.15 μg/mL.
Compound 19-04-B-F8 requires additional spectroscopic analysis and purification
of the active in order to establish it as a α-glucosidase inhibitor.
