Isolasi Dan Uji Antidiabetes Senyawa Metabolit Sekunder Dari Ekstrak Diklorometana Tumbuhan Paku Pyrrosia Longifolia

Pyrrosia longifolia is an epiphytic plant in the Polypodiaceae family. This species
grows on large and old trees, typically on trunks, branches or twigs. P. longifolia
study is still underreported, particullary its bioactivity, whereas secondary
metabolite isolation and bioactivity as an antioxidant, anti-inflammatory and
antidiabetic have been reported in other species such as P. petiolosa, P. sheareri
and P. lingua. This aims of this research were to isolate secondary metabolites
from the dichloromethane aerial parts of P. longifolia extract and to eveluate their
antidiabetic activity. P. longifolia was macerated with methanol as the solvent and
partitioned with various solvents (n-hexane, dichloromethane, and ethyl acetate).
Using liquid vacuum chromatography, the dichloromethane extract was purified
into 17 fractions. In fraction 3, crystalline solids were found, and recrystallization
was perfomed to obtain pure crystals, which were coded as samples 20-05-B-F3.
Thin layer chromatography (TLC) and a melting point of 58-60oC were used to
determine the purity of 20-05-B-F3. The compound characterization using FTIR
spectroscopy revaled the presence of aliphatic CH bond vibrations at wave
number 2916 cm-1, C=O ester groups at 1734 cm-1, CH2 bond vibrations at
1472 cm-1 and bending vibration CH at 1375 cm-1. Based on percentage of
α-glucosidase inhibition from P. longifolia extracts, n-hexane, dichloromethane,
ethyl acetate, and water extracst showed weak inhibition with value of 7.5 %,
16.2 %,6.8 %, and 20.4 %, respectively at concentration of 500 ppm.
