Hemorrhoid is anorectal disorder characterized by venous dilation, inflammation, and structural damage to the supporting tissues of the anus. Inflammatory mediators such as COX-2 (pro-inflammatory) and IL-10 (anti-inflammatory) play key roles in their pathogenesis and serve as biomarkers for disease progression and therapeutic response. Curcumin, the active compound in turmeric, has known anti-inflammatory properties through regulation of COX-2 and IL-10 expression; however, its low bioavailability limits clinical use. To overcome this, nano curcumin was developed to enhance absorption and stability. This study aims to analyze the effects of nano curcumin on COX-2 and IL-10 expression in a hemorrhoid rat model in vivo.
Methods: This study utilized an experimental laboratory design with a rat model of induced hemorrhoids. The rats were divided into five groups: a negative control group, a positive control group, and groups receiving nano-curcumin at doses of 80 mg/kg and 160 mg/kg body weight. The expression of COX-2 and IL-10 was evaluated using immunohistochemistry and RT-PCR to assess the inflammatory response following nano-curcumin administration.
Results: The administration of nano-curcumin at both 80 mg/kgBW and 160 mg/kgBW significantly reduced the expression of COX-2 protein and mRNA in hemorrhoidal tissue (p < 0.05), indicating a reduction in the inflammatory response. Furthermore, nano-curcumin treatment significantly increased the expression of IL-10 protein and mRNA (p < 0.05), suggesting anti-inflammatory and immunomodulatory effects that contribute to tissue repair.
Discussion: Nano-curcumin has the potential to be an effective anti-inflammatory agent by reducing COX-2 expression and increasing IL-10 expression in hemorrhoids. This study provides scientific evidence supporting the development of nano-curcumin-based alternative therapies for managing hemorrhoids.
